Sengen, There are three mechanisms could be used in reaction selection. One is Acidic-based demethylation, another is Basic-based, and the last one is Reductive demethylation. In your case, I think Basic condition might be the best choice, like NaOCH3, NaCN/DMSO, or NaSPh/HMPT. Demthylation is one of the most difficult reactions I ever face in my 15 year Med. Chem. experience. Good Luck!
