James:

Is there a possibility that your aldehyde analytes have oxidized on the
plate to a greater or lesser extent before you try to detect them? I
have known aldehydes to go quite quickly - 30 min or less from the time
of application.

Check your stains by testing them on spots of the aldehyde BEFORE
developing the plate to ensure that they work. If you get good
intensity, then fine. Next, try developing freshly spotted samples. If
you don't get similar intensity after developing (allowing for lateral
diffusion), then oxidation during development, or between development
and staining is the most likely problem.

Subject: ORGLIST: Stains

Hi all,
I am currently working with Piperonal and
3,4-dihydroxybenzaldehyde and ultimately 3,4-dihydroxyacetaldehyde and
other similar aldehydes. I want to make sure that these components have
reacted fully by staining them on TLC plates but so far my permanganate
stain and anisaldehyde stain do not seem to be working as effectively as
they should. I believe that they were prepared correctly as I prepared
them from this link
<http://www.chem.rochester.edu/%257Envd/tlcnotes.html> . Can anyone
suggest a better stain? Thanks.
